Hi can I use the Duet 3 with SBC (RaspberryPi) and a connection via USB with another PC simultaneously?

I'm not sure... But if you do try it, be mindful of USB ground loops.

@rabbit yes that should work, but don't try to do send a print file through USB because the Pi won't know that you are doing a print from file. And be careful of ground loops.
