Thank you both! you were correct and it solved my problem printer is up and running. Very much appreciate you guys helping me out with this and information that will indeed be useful in the future.

You just need to map

+5v -> 5V_EXT
GND -> GND
URXD0 -> IO0_0.out
UTXD0 -> IO0_0.in

Just like shown on the wiki https://duet3d.dozuki.com/Wiki/Getting_Started_With_Duet_3#Section_Running_in_standalone_mode

It works.
The issue was the copy I initially downloaded, it might have been corrupted. Everything looked good [write, verify] up to the reset point, the board stopped communicating.
